Karolinska Developments portföljbolag AnaCardios grundare publicerar studie som stöder utveckling av AC01 för behandling av hjärtsvikt

Karolinska Development AB (Nasdaq Stockholm: KDEV) meddelar idag att portföljbolaget AnaCardios grundare, professor Lars Lund, har publicerat en studie inom hjärtsvikt i den vetenskapliga tidskriften European Heart Journal. Publikationen av studien stärker bolaget i sitt arbete med att utveckla en helt ny typ av behandling av hjärtsvikt baserat på proof-of-concept som genererades i studien.

Publikationen i den vetenskapliga tidskriften European Heart Journal baseras på en studie som leddes av professor Lars Lund och genomfördes i samarbete med forskare vid Karolinska Institutet och Karolinska Universitetssjukhuset. Studien utvärderade peptidhormonet ghrelin som behandling hos patienter med hjärtsvikt samt i isolerade hjärtceller, och visade på en ökad sammandragningsförmåga och en ny behandlingsmekanism.

Studien var randomiserad, dubbelblind och placebokontrollerad och omfattade 30 patienter med hjärtsvikt. Patienterna randomiserades till att antingen få intravenös behandling med aktiv substans eller placebo under två timmar. Behandlingen med ghrelin resulterade i att den volym blod som pumpas ut av hjärtat ökade med i genomsnitt 28 procent i ghrelingruppen, medan placebo inte gav någon effekt på pumpförmågan. Inga allvarliga biverkningar observerades och inte heller några tecken på hjärtrytmrubbningar, syrebrist i vävnaden eller lågt blodtryck. De underliggande mekanismerna som låg bakom den ökade pumpförmågan utvärderades och identifierades i prekliniska modeller på aktiva hjärtceller i laboratoriemiljö.

Baserat på denna väldokumenterade verkningsmekanism har AnaCardio initierat en klinisk studie för att utvärdera läkemedelskandidaten AC01, som är en småmolekyl som härmar ghrelins verkningsmekanism och har god potential att på ett säkert sätt kunna förbättra hjärtats sammandragsningsförmåga hos patienter med hjärtsvikt.
